Description
- Recognizes human CXCL10
- CXCL10 (IP-10) is a member of CXC chemokine family that binds to CXCR3 receptor to exert its biological effects
- 12-kDa protein, constitutes and constitutes two internal disulfide cross bridges
- Cleavage generates a 10-kDa secreted polypeptide with four conserved cysteine residues in N-terminal
- CXCL10 mediates lekocyte trafficking, adaptive immunity, inflammation, haematopoiesis and angiogenesis
Process:
- Antibody specific for CXCL10 pre-coated onto microwells
- CXCL10 protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody of biotinylated specific for CXCL20 is added to detect captured CXCL10 protein
- For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
