Description
Streamline antibody-drug conjugate screening campaigns with Invitrogen Zenon pHrodo IgG Labeling Reagents to screen for antibody internalization. These reagents provide rapid, scalable antibody labeling and give a bright fluorescence-based indication of antibody uptake and trafficking in target cells when analyzed by flow cytometry, fluorescence microscopy, or high-content screening.
Zenon™ pHrodo™ iFL Green-labeled or Red-labeled Fab fragments (i.e. labeling reagent) are directed against the Fc portion of an intact IgG primary antibody to form a labeling complex. Formation of the Fab–antibody complex occurs in fewer than 5 minutes. Because the pHrodo™ iFL dyes dramatically increase in fluorescence as the pH of their surroundings becomes more acidic, the antibodies coupled with the Zenon™ pHrodo™ iFL IgG Labeling Reagents provide an excellent measure of endocytic activity based on the acidification of the labeled antibodies as they are ingested by the cell.
The Zenon™ pHrodo™ iFL IgG Labeling Reagents contain sufficient material to label one to four 96-well plates, depending on the Zenon™ labeling reagent concentration used. Only a small amount of starting material is needed, and the method is optimized for efficient labeling of antibodies in serum, ascites fluid, or hybridoma suspensions.
Other advantages of the pH-sensitive Zenon™ pHrodo™ iFL IgG Labeling Reagents include:
- Simplicity—No reagent purification is required
- Accuracy—No wash steps or quenchers are needed, minimizing cross-reactivity or background
- Speed—Labeled antibodies are typically ready to use in 10 minutes
- Specificity—Dyes are designed for the specific detection of phagocytosis and endocytosis
Preserve primary antibody function and affinities
Reactive dye labeling of primary antibodies can have unpredictable and undesirable outcomes. One of these is reduced binding affinity due to label addition in the binding pocket. The Zenon™ antibody labeling approach, which is targeted to the Fc tail, resolves this potential issue.
