Description

Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous PCSK9

  • Recognizes human PCSK9
  • PCSK9 (NARC1, PC9) belongs to peptidase S8 family
    • Expressed and secreted by tissues (liver, small intestines, kidney)
    • May be implicated in differentiation of cortical neurons, may play role in cholesterol homeostasis
    • Regulator of LDL receptor and plasma cholesterol concentrations

Process:

  • Antibody specific for PCSK9 pre-coated onto microwells
  • PCSK9 protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody of biotinylated specific for PCSK9 is added to detect captured PCSK9 protein
  • For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
