Description
- Recognizes human MMP-9
- MMP-9 (Matrix metallopeptidase 9, Gelatinase B) is a member of matrix metalloproteinase family
- MMP family of enzymes comprised of critically important extracellular matrix remodeling proteases whose activity implicated in normal embryogenesis, tissue remodelling and diseases (arthritis, cancer, periodontitis, glomerulonephritis, encephalomyelitis, atherosclerosis and tissue ulceration)
- Proteases represent important therapeutic and diagnostic targets for treatment and detection of human cancers
- Produced by variety of normal and transformed cells (neutrophils, monocytes, macrophages, astrocytes, fibroblasts, osteoclasts)
- Destroys collagen (Type IV) in vascular basal membrane in vicinity of tumor cells which invade surrounding tissues and contribute to metastasis
- Circulating levels increased in many inflammatory disorders (atherosclerosis, hepatitis C virus infection, colorectal cancer)

Process:
- Antibody specific for MMP-9 pre-coated onto microwells
- MMP-9 protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody of biotinylated specific for human MMP-9 is added to detect captured human MMP-9 protein
- For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
