Solid phase sandwich ELISA used to detect and quantify protein levels of endogenous gp130

  • Recognizes human gp130
  • gp130 (Glycoprotein 130, IL6ST, CD130, IL6-β) is ubiquitously expressed, signal-transducing receptor that serves as signal transduction unti for IL-6 family of cytokines (IL-6, IL-11, IL-27, LIF, OSM, CNTF, CT-1, CLC)
    • Cytokines signal through gp130/Jak/STAT pathway
    • Binding of IL-6 to IL6R induces gp130 homodimerization and formation of high-affinity receptor complex, which activates Jaks
    • Causes phosyphorylation of gp130 tyrosine residues which activates STAT3
    • gp130 is a Type I transmembrane protein
      • Soluble form (sgp130) binds to sIL-6R/IL-6 complexes and prevents interactions with membrane-anchored gp130 on target cells

Process:

  • Antibody specific for gp130 pre-coated onto microwells
  • gp130 protein in samples captured by the coated antibody after incubation
  • After extensive washing, another antibody of biotinylated specific for human gp130 is added to detect captured human gp130 protein
  • For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
  • Solution containing sulfuric acid used to stop color development and color intensity
    • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
