Description
- Recognizes human APOB
- Apoloprotein B (APOB) is a plasma protein synthesizes primarily in the liver
- Essential for assembly of VLDL in liver and intestine
- Plays role in lipid and cholesterol metabolism
- APOB gene encodes two isoproteins through mRNA editing (APOB-48 and APOB-100)
- APOB-100: synthesizes by liver and essential for assembly of VLDL in liver
- APOB-48: synthesizes by small intestine and essential for assembly of chylomicrons in intestine
- Plasma APOB levels associated with coronary heart disease
Process:
- Antibody specific for APOB pre-coated onto microwells
- APOB protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody of biotinylated specific for human APOB is added to detect captured human APOB protein
- For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
