Description
- Recognizes human AFP
- AFP (alpha-fetoprotein) is a major plasma protein found in fetus while plasma levels decrease rapidly after birth
- Early marker of hepatocyte lineage
- High concentrations correlated with tumor cell growth
- Detection of AFP important to diagnose hepatocellular carcinoma, stomach cancer and germ cell cancers
- Altered levels of fetal and maternal AFP associated with hypothyroidism, autoimmune disorders and heart defects
Process:
- Antibody specific for AFP pre-coated onto microwells
- AFP protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody of biotinylated specific for AFP is added to detect captured AFP protein
- For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
