Description
- Recognizes human ABCD1
- ABCD1 known as adrenoleukodystrophy protein (ALDP)
- Member of multi-pass membrane protein
- Localized at peroxisome membrane
- Mutations of ABCD1 cause X-ALD, an inherited neurodegenerative disease affecting the nervous system, white matter and adrenal cortex
- Results in mental deterioration, corticospinal tract dysfunction and cortical blindness
Process:
- Antibody specific for ABCD1 pre-coated onto microwells
- ABCD1 protein in samples captured by the coated antibody after incubation
- After extensive washing, another antibody specific ABCD1 is added to detect captured ABCD1 protein
- For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
- Solution containing sulfuric acid used to stop color development and color intensity
- Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
